HPLC verification refers to analyzing a substance through High-Performance Liquid Chromatography to verify its precise chemical identity and assess its purity percentage.

What It Implies

  • Identity Confirmation: Assesses the duration it takes for a compound to navigate the system (retention time) compared to a known reference standard.

  • Purity Percentage: Evaluates the proportion of the target peak to the total peaks displayed on a graph (chromatogram), frequently indicating purity levels such as ≥ 98%.

The Verification Process

  1. Sample Preparation: Dissolve the solid or liquid sample in an appropriate solvent and filter it to eliminate particles that may block the instrument.

  2. Injection: Introduce a small volume of the prepared sample into the HPLC apparatus.

  3. Separation: A high-pressure pump forces a liquid solvent (mobile phase) through a densely packed tube (column). Various molecules travel at varying speeds depending on their chemical characteristics.

  4. Detection: As compounds exit the column, a UV (ultraviolet) or photodiode array detector measures them and converts the results into a visual chart of peaks.

  5. Data Analysis: Software computes the area under the primary peak in comparison to secondary impurity peaks to generate a final purity score.
